Development of species specific PCR marker for the detection of Phytophthora capsici from RAPD marker

A species-specific PCR marker has been developed for rapid and accurate detection of Phytophthora capsici from diseased plants and soil. The marker was generated from the RAPD marker obtained on genetic diversity studies of Phytophthora capsici isolates. The specific band (~550bp) obtained with primer OPAA10 were cloned sequenced and converted into a sequence characterized amplified regions (SCAR) marker. The SCAR primer amplified a 500bp amplicons from Phytophthora capsici isolates of black pepper, capsicum, cocoa and nutmeg. The specific amplification of SCAR primer was highly sensitive, detecting template DNA as low as 10ng. The SCAR amplified the specific product from diseased host and soil DNA samples. The results indicated the potential of SCAR primers for detection of P. capsici from the environmental samples.
